THERAPEUTIC EFFECT OF D-PENICILLAMINE AND CYCLOPHOSPHAMIDE IN PARAQUAT POISONING : SURVIVAL RATE IN RABBIT FOLLOWING SUBCUTANEOUS INJECTION.

ABSTRACT
Paraquat(1.1-dimethyl-4,4-bipyridium dichloride) has toxic effect by turing into a stable free radical in the presence of powerful reducing agents. Among the complications of multiple organ failure, the pulmonary fibrosis is always fatal. Immunosuppressive therapy with cyclophosphamide suppress the formation of oxygen free radical, superoxide, and hydrogen peroxide. D-peniciallamine inhibits the formation of collagen cross-link by blocking aldehyde groups on synthesized collagen molecules. Therapeutic effect of cyclophosphamide and D-penicillamine was studied on 30 rabbits. Thirty rabbit were allocated into 3 groups, each group consisting of 5 males and 5 females, which were injected subcutaneously with 10mg/kg of 20% Gramoxone. The control group(n=10) had no treated, the group I(n=10) had treated with cyclophosphomide(5mg/kg/d) and the group II(n=10) had treated with D-penicillamine(40mg/kg/d) and dexamethasone(1mg/d) The urines were collected by nelaton catherization 2 hours after injection and the qualitative test was done using sodium dithionite. The 80% in control and group I and 70% in group II were showed strong positive in urine color. The urine test showed 100 % proteinuria in all groups. The 5 rabbits were died and the total survival rate was 83%. The survival rate was 70% in control group and 90% in both experimental groups. The mortality rate according to sex was male 4/15(26.7%) and female 1/15(6.7%). On the pathologic finding of lung, there was a no specific difference in control, experimental group I and group II which were survived at 2 weeks later. But the alveolar rupture and severe emphysematous change were found in 3 died rabbits's lung of control group. Our results show that cyclophosphamide and D-penicillamine have improved the survival rate in the rabbits with paraquat intoxication. But there are no defference of survival rate and pathologic findings between the experimental groups.
